Checking the "sealed" transmission behind the 4.0 is similar to checking a manual transmission. On the transmission housing under the truck, there is a fill plug on the top, check plug on the side, and drain plug on the bottom.
The transmission must be at the proper temperature, and when it is, remove the check plug and confirm that the fluid is at the edge of the hole.
But unless it is leaking, the amount of fluid will be the same as when you bought the truck unless the cooler in the radiator is leaking (check coolant for oil).
Toyota considers the sealed transmission to be a "lifetime" fluid, but the dealership will be happy to change your fluid if you would prefer to do so. I recommend every 100k unless you are towing.

Trans fluid goes in the same tube the transmission dipstick sticks into. You'll need a funnel with a narrow bottom opening (1/2" or less). Make sure you pour it in the transmission dipstick tube, and not the engine oil dipstick tube. ;)
Initially, try to add about 3/4 of the estimated amount that you accidentally drained, then slow fill the remainder while checking level at different stages.
Also, try not to overfill. Check fluid level often, with engine running, in park or neutral, as you re-fill it.

you put ion the amount to bring back to the correct level on the dip stick
unless you have completely drained the transmission AND the torque converter then the amount required is around 1/2 the capacity stated in the book
so start with the 2.1 quarts monitoring the dip stick until you get it on the mark then follow the instructions in the user manual as to starting /running in gear at idle and topping it off as required

if there is no dip stick then it must be a stick. if it is the should be a drain and fill plugs on the trans. the top one is the fill. undo the plug and if you can touch the fluid with your finger there is enough fluid in the trans

your fluid is low. to check the fluid make sure your car is running and check the stick a few times wiping it off between dips. DO NOT overfill the fluid and make sure you use the right fluid for your tranny- ask for the right fluid in your local kragen etc.

You need to find out were the fluid is leaking from first...
It may have been because the the dip stick was half way out...
Id just keep an eye on the fluid levels and if it stays the same than that's what it could have been.
