SOURCE: Tuono R 2007 Engine cuts out when fan kicks in and won;t

By fan you mean the radiator fan? If so try the following. If a coolant temperature of between 239oF(115oC) and 275oF(135oC) appears on the multifunctional digital display, stop the vehicle and leave the engine running at 3000 rpm for about 2 minutes, allowing the coolant to circulate regularly in the system. Then position the ENGINE STOP switch to "OFF" and check the coolant level to make sure it is ok. If after checking that everything is ok it still does the same thing the you should probably go to a mechanic.

My kick starter just locked up

1). Confirm that the transmission is really in nuetral, or insure that the clutch is pulled in.

2). Have you tried removing the spark plug to insure that you do not have a hydraulic lock due to excess fuel in the cylinder? With spark plug removed, any excess fuel should be expelled out the open spark plug hole. CAUTION: INSURE THAT THE SPARK PLUG LEAD IS DISCONNECTED, AND HELD AWAY FROM THE OPEN CYLINDER TO PREVENT ACCIDENTAL IGNITION OF EXCESS FUEL.

My Yamaha yz250F 07 starts first kick but back

I see: you changed the plug because it was backfiring.

Does it back fire when you let OFF the throttle after revving it, or does it back fire while being revved?

If it does it when you let off then Ollie's suggeston might do it. If that doesn't do it then check for dirt in your pilot jet.

If it back fires when revved up high could be dirt in your main jet.
