Try replacing the spark plug and the spark plug cap. If the resister in the spark plug cap burns out it will cause a weak or no spark. Most plug caps simply unscrew counterclockwise off of the end of the plug wire. An NGK replacement plug cap will cost between $5 - $10. With the cap off the wire you can check the resistance with a volt meter. Most caps should read 5000 OHMs , any reading higher or lower means it is time to replace the bad cap.

If you have spark than, check for fuel flow from the petcock to the carburetor. If fuel is flowing to the carburetor than try disassembling and cleaning clean the carburetor.

The carburetor is actually two with a common air intake and fuel bowl yet separate jets and and manifolds to each cylinder.

Disconnect plug wires and crank for a few seconds. Promptly remove both spark plugs and compare. Likely one will be wet (good cylinder) and the other dry (bad cylinder).

If both plugs are wet then could be no spark under compression.

What does this mean cylinder 2 misfire

It means that #2 cylinder is not running right. It could be a bad spark plug, bad spark plug wire, bad injector, or bad coil pack. You will need to find out if you have spark on that cylinder. If you have spark, then you will have to check to see if it is getting fuel properly. This could also include a bad valve in the engine. If the compression is not right the cylinder will not fire either. I hope this helps you.

Why does only 1 exhaust pipe on my ninja 250 get hot

Try unplugging one spark plug at a time and see if the engine will run on just left and just right cylinder. The cylinder thatwont run may have a clogged carburator or the valves may be too tight causing a low/no compression on that cyliner. Also a vacuum leak at an intake boot would cause a lean condition and a hot cylinder. Be certain you have spark at both ( properly gapped) spark plugs before **** these tests.

Check distributor cap for cracks

Run a compression test on all cylinders. +/- 10% across all cylinders and upwards of 140psi.

Change HT leads. Make sure you get them in the right order.

Not running on #3 cyl' on my 1982 gs550

Not likely valve timing if it will run, cams are one piece so if one is right all are right. Have you checked compression with a gauge, or just by feel? Try swapping the sparkplug wire with the #1 cylinder and see if the dead cylinder moves with the wire. Also try moving the spark plug to another cylinder and see if the dead cylinder moves with it. If the compression is indeed good, the problem will be with the spark plug/ wire, or in the carb, these are the only things that can kill a single cylinder. The coils run a pair of cylinders so a bad coil would put out 2 cylinders at the same time. Start with the spark plug, I have seen a perfectly good looking plug, that would fire whun held to the head, but would not fire in the cylinder.

You need to check spark plug wire to that cylinder I have had the same problem where a mouse ate insulation off wire ,Your engine has a coil with two wires one runs each cylinder I have had them go bad where only one fired you would need a new coil .
