I have a2008 GR650S and had the same problem. Bought the secondary air injection kit (see link) and the problem as solved. However it does affect the anti-polution compliance for the bike. Fitting the blanking plate to the rear cylinder is difficult.

Mate i had the same issue. Just got my first 1000km service. I had a leak in the fuel line causing some petrol to get into the exhaust. common problem but easy fixed. was told the line just cracks sometimes.
Do you have a aftermarket exhaust? If so then your bike probably wasn't re jetted after the exhaust was installed. A rejet kit is like 50 bucks, try that.
Backfires are when fuel travels through the cylinders without combusting then explodes in the exhaust pipe. Fuel supply/combustion issue. It seems likely there's a problem either with the mix going into the cylinders (carbs fuel lines or air intake), or with timing. These can affect a bike at particular speeds and throttle positions. If using the choke has helped at some point that's a further indication (the choke changes the gas/air mix). ,,,
Go to a mechanic because the fuel-to-air mixture is 'rich' that's why it backfires
