I have a 2001 TTR225 and just had the same issue. I had not riiden the bike in a while. Fuel was flowing through the petcock and fuel line. I removed the carb bowl and cleaned it and the jet. Didn't work--still no fuel in the bowl. Ultimately I had to remove the the bowl, floats, and the needle connected to the floats. The needle conected to the float and brass cylinder that it is in were gummed up. Be careful, the floats are delicate and the tiny retainer spring is...tiny, but tough to bend. Slip the spring over the float arm, then remove the little philips screw below it. The needle and brass cylinder will slide out with a needle nose pliers. Reassemble by reconnecting the needle to the float and slide in as a one assembly. Good luck.
Adjust your valves
Adjust the fuel screw which is under the carb
Remove fuel line from petcock and see if fuel runs out, if not remove petcock from tank and clean filter screen, if it does come out, check for inline fuel filter change it, if it dont have one check fuel pump if it has one, or could be diaphrams in carb split and not building vaccuum to draw gas thru
