There could be many reasons for this, some simple, some not so simple, so you will need to start by eliminating simple things first.
The first thing to check is the battery. Is it fully charged and in good enough shape to supply the electrical load needed to make the bike run correctly (bikes with electronic ignition and fuel injection must have a good battery for the bike to run properly)?
Next, take a look at your owners manual and find out if there is a fuse for the fuel injection system. If there is, is it good?
When you insert the key and turn the ignition on can you hear the fuel pump? If not, you fuel pump is either dead or not getting power for one reason or another.
Many cars are equiped with a safety switch which automaticly turns off the fuel pump in the event of an accident. These can be reset by simply pushing a button. Is your bike equipped with something similar?
When was the last time the bike was running? The gas sold today is notorious for not being stable for very long. If everything else on the bike checks out ok, there is a remote chance your injectors are clogged with bad fuel.
the float bowl evaporates in a little over a week .
When the system has some movement added to it ,the bowl will re-fill.If it were fuel injected the fuel pump would add pressure to the fuel system right away.
There might be an after market fuel pump that is electric that might solve your problem all together.

Before I started "opening it up", I'd try to determine why the front cylinder is not running. You gave me absolutely nothing to work with here other than it has been sitting for a "few weeks". Specifically how long is a "few weeks". Even though the gasoline we buy these days is pretty poor quality, it will still last more than a few weeks unless that few weeks is more like a few months. Usually an engine does not break when it's not running but there are a few things that can go wrong. A tappet could collapse and then stick causing a loud noise but even with a collapsed tappet, I've seen them run. You could have a spark plug fouled and not firing. Since Harley uses a dual fire type ignition where both plugs fire every time the pistons come to top dead center, if one plug fires, then the other should be firing. That covers the ignition and compression, leaving only fuel. You failed to say if the engine was equipped with a carburetor or fuel injected. Since it's missing only on one cylinder I have the tendency to think that it may be fuel injected. If so, that's where I'd look to begin with. If it's a carbureted engine, look for a vacuum leak somewhere feeding the missing cylinder.
Do not touch the throttle till it fires. Only the choke, which is the primer for the start up fuel. The throttle does nothing till the engine is running. Your problem sounds like a weak battery. If it is like mine, I have been wrestling with a slow starter for years, the battery is tiny. And even a brand new one is slow. I have replaced the battery 3 times in 8 years. I know you can check the pressure of the fuel pump, but if it is humming, it should be working. It comes on with the key and it stays on. Don't forget, you are fuel injected, 30lbs pressure, min, or something like that. Hope this helps, 2002 S4, 1999 750 monster.
Bike should be fuel injected so very hard to flood. Most likely battery was weak from cold and couldn't turn over and spark enough to start. Try charging battery for a few hours, beware of sparks if you smell gas. Hook charger up then turn on.
First, I'd check the battery. Just because it's new doesn't necessarily mean anything. I've seen them bad right out of the box. Make sure you've at least 250 cold cranking amps in the battery.
Next, find out why the bike doesn't start on the first try. You said "after a few goes". You're using a might small battery to start a large engine. It can't turn in over forever. If you have a carburated engine, turn on fuel petcock, pull out the enricher, milk the throttle twice, turn on the ignition switch, press the starter button. Do Not open the throttle at all. If you do, you disable the enricher circuit. If this doesn't work, check to make sure your accellerator pump is working correctly.
If your bike is fuel injected, a trip to the deal is warranted. No user adjustable parts there.
Is the bike stock? If you bought the bike used, there's not telling what a previous owner may have done to it in the line of high performance work. You may want to run a compression test on the engine to determine if that cams or anything else has be replaced. If so, buy the biggest baddest battery you can squeeze into the bike. Harley's AGM battery is a good battery for a stock or relatively mildly modified bike.
Didn't say much about what it does or doesn't do as trying to start. The batrtey all the way up? if not it can refuse to start even if cranking 2002 and newer may push start but only if the battery is near good 04 and newer are fuel injection and a low or bad battery is a no start on it's own the modern glass mat batteries don't take a jump very well as they resist being pushed back to full voltage even with a car battery. Cranks good with a good battery. Got gas and the valve on? Carb bikes may have to have the fuel pet on prime to start a (dry) motor. The fuel injection bikes have to have the good battery or the fuel pump can be below theasehold and the relay won't turn the pump on. All the lights and others horn ect working? No crank can be clutch switch kickstand switch if the locic box is burnt out or low voltage, Neutral light working? if not the kickstand has to be up for crank. Burn the strarter out cranking with low battery? Strong click when pushing the start button says the safty switches are working. and the starter could be bad or lost it's ground. Will the engin turn? put the bike in higher gear and push it with the clutch out to see if it will turn good compression may be hard to push but not impossible as the starter and you have about the same power, remember they used to have kickers and this one had one it'd work to start it. Carb bikes have drain bolts on the float bowels and little hses hooked to them if their still there open the screws to see that theres gas or something else in the corbs. and will more come from the tank? Spark the plug wires snap to the plugs and are hard to remove if good put a plug ( any good plug will do) and crank the engine to see the spark at the wire. if not kill switches crash swiches may have to be reset then the key on and off too. lfter that its lookto see whats missing as compression spark and fuel are all it needs to run. If it jumped time the compression won't be good or out rajous. Exhaust stopped up? I've seena mouse store corn in the exhaust and forget wher he packed it. seen a R6 completly full! no start air cleaner housings get filled the same way. these don't apply if the bike had been running recently. Bad or poor battery most likly cause of no start then cable connections
from your description, i would sugest you make sure fuel is getting to the injector unit, remove the fuel supply hose.if you have spark at the plugs and fuel getting to the injectors you have isolated the problem. i would also drain the old fuel when you take the hose off and fill the tank with high grade fuel. bikes dont like old gas.
