It depends on whether the sprockets are original size or not. Counting links doesn't work unless you are positive some previous owner did not take links out of the chain.

The lenght of the chain is de noted by the amount of inner drive links it has, there should also be some information etched into the bar, usualy a number follewed by DL ( drive links )
Impossible to tell you without knowing exactly what your chainstay length is, the rear cassette your using, the rear derailleur your using and the cranks your using and even then could be a wrong answer. With chains you have to measure the amount you need by installing the chain and seeing how much excess there is for your configuration.
you said it all, most bikes have a master link, if not you need a chain breaker, which you can also use if the new chain you purchase is to long. lift bike on center stand, loosen axle nut, then loosen chain adjustment screw nuts on both sides and turn them so the wheel/axle moves all the way forward. connect the old chain to the new chain and pull it through (while gearbox is in neutral). adjust the chain adjust screws on the axle the same amount indicated by the markers on swing arm until chain has a deflection of half inch up and down in the middle at the tightest spot, try turning the wheel a few cycles to find it. then lock axle nut then tensioner lock nuts.
when you put the chain on new did you adjust the tension on the chain.most new chains need to be ran a few minutes to heat them up a little.then you need to readjust chian so it doesn't sag out of the bottem of the bar,also when you adjust the chain you need to push the tip of the bar down so the bar is in the up possition.if you don't the bar will work it's way up and loosen the chain.
