I have a canon EOS 40D, and suddenly it stop working. When I press the shutter button half way, the auto focus works but the camera is not taking the photo when I completely press the shutter button. I have not had this problem since I bought this camera about a year ago.
I have the same problem and it happened whilst I was taking photos this morning - auto focus works as does info screen (shutter speed/app etc..) on top of camera. However the camera will not take a photo and the LCD screen on the back does not come on - cannot get it to playback photos from memory and it does not even display the "cleaning sensor" graphic" when turned off. when taking a picture with the AF set the buton depresses half way but wont fire, however on MF it does work. When the dial is set on 'M' iget the full menu but on auto I get a much more limited menu

Try removing the lens with the camera off and re-mounting it. Before doing so, check to make sure the contacts on the lens and camera body are clean and not damaged. If that doesn't help try gently pressing on the front of the lens (not the glass) while pushing the shutter release button half way down and if it works then you've got a poor fit between the lens and camera body.
3) Canon had a run of bad motors for this model - if mirror does not go up, then it is the mirror motor. If mirror goes up, shutter will not fire, press release again then the mirror goes down, the shutter motor is bad (same model motor). If mirror goes up, shutter fires, both blades travel together/no exposure, probably the shutter is bad.
Cannon is notorious for their auto focus and automatic setting to be very discriminant. If you are to close to your subject while in these settings granted your strobe will fire and your flash may flip into position but the shutter just wont work. Cannon is trying to tell you you are too close. You need to back up. If you are trying to get that close up and don't have the proper lens that's all you are going to get. It is best to go long lens then go for your close up.
This is just a possibility not really a solution: If the AF spot is aimed at a part of the picture lacking details, it cannot focus (all cameras are the same in that respect) Normally (I don't know the canon) there is an indicator to tell you if the camera managed to focus. It can be a beep or a dot which light in the viewfinder. I suspect that in your case you were aiming at something lacking details.
That is a custom function of the camera that can be change back to default with ease. While in one of the creative zones press the menu button and navigate to the 5th and final tab on the top right.
This is the Setup menu 2. In this menu, select “Custom Functions (C. Fn)” . This will open a submenu of numbered options. Select #04 and set it to 0: the default AF/AE lock on a half depress of the shutter.
That should get your camera back to the expected auto focus with the shutter button.
Sounds like your custom functions have been changed to work this way. On the EOS 350 D it is Custom Function #04. To change it back to default you need navigate through Canon's menus.
