All of the sudden today on the above remote, there is a 'S' inside a box under the lock/unlock icon on the display. I can't find out what it is on any of my documentation.
That little "S" is the power save feature. Press the "F" and the lock buttons together to toggle the feature on and off. With the S on the battery in the transmitter will go into a power down mode when alarm is in disarmed state to save battery life.

OPEN Driver’s door and INSERT key into the Ignition.TURN the Ignition to ACC (one turn back towards you) and then to the LOCK position three (3) times within three (3) seconds.Within 10 seconds, CLOSE and OPEN the Driver’s door twice (2).AGAIN, TURN the Ignition to ACC and then to the LOCK position three (3) times within three (3) seconds.AGAIN, within 10 seconds, CLOSE and OPEN the Driver’s door once (1). The vehicle will respond by cycling the locks indicating that the vehicle has entered Programming Mode.Within 20 seconds, PRESS the LOCK Button on the remote to be programmed. The vehicle will respond by cycling the locks.Within 20 seconds, PRESS the UNLOCK Button on the remote to be programmed. The vehicle will respond by cycling the locks signalling successful programming of the remote.REPEAT Steps 7 and 8 for each additional remote to be programmed to the vehicle.REMOVE the key from the Ignition to exit Programming Mode.Test all remotes. There are two possiblities that could prove to be the fault; hood pin switch and remote start shutdown switch. If the vehicle is equipped with a hood shut down switch, you will need to make an adjustment to it for the remote starter to function. Search around the drivers side edges of the engine compartment for a push pin.If the pin is a stand alone it will be mounted near the headlight inserted into an existing hole. It may also be attached to a bracket. The remote start shutdown switch is a toggle switch located under the drivers side of the dash,usually to the left of the brake pedal, depending on the installation.
If nothing above solves the problem, you may need to reprogram the remote to the unit.

both the rear passenger side wont unlock ???? How many doors are on the passenger side ? Or both back doors don't unlock ?
Two relays lock / unlock the back door's Not the lift gate . The BCM controls the relay's . The relay's are located in the rear fuse box . Under back seat .
The lock/unlock outputs from the main unit are reversed. There are two negative output wires from the unit that control the doorlocks, if you just switch them around, they will function correctly.
