If you have a carrier locked iPhone (AT&T in the US), then simply connect the iPhone to your computer with the iPhone's USB cable and open iTunes. Upon doing so, iTunes should connect to the internet and activate your iPhone by confirming your AT&T service account via Apple's activation servers.
If your iPhone is using a Non-AT&T carrier, then you must download and use an application like Blackra1n to re-activate your iPhone and jailbreak it, if necessary.
However, please note, if you need the iPhone unlocked as well, firmware OS version 3.1.3 (currently the latest, as of this writing) is not unlockable due to the changes it makes to your baseband. You cannot downgrade your firmware OS either since the baseband changes 3.1.3 makes to your iPhone's modem are permanent.
Until a proper fix is found and released by the iPhone Dev-Team (the creators & supporters of the iPhone unlocking software), the best you can hope for is an activated, jailbroken iPhone, but sadly, not unlocked.
